HPLC/UPLC spectrophotometry for Formazan dye quantification in in vitro tests
What if your product is of a strongly colored nature in a matter that it can’t be properly tested in an MTT reduction-based assay? This is the case when the substance itself absorbs light in the same wavelength range as the evolving formazan dye (FD).
FD is the key marker for cell viability and tissue integrity in OECD 439 (In vitro Skin Irritation) and OECD 492B (In vitro test method for Eye Hazard identification).
At LAUS, we have qualified the HPLC/UPLC spectrophotometry procedure described in the OECD TG 492B. That allows reliable quantification of FD and a meaningful and compliant evaluation for strongly colored test items.
