Trouble with strongly colored test items in an MTT assay like OECD 492B?

In vitro Eye Hazard Identification can get tricky when your test item is strongly colored and absorbs light in the same wavelength range as the formazan dye (FD) – the key marker for cell viability and tissue integrity.

At LAUS, we are able to help:
We have qualified the HPLC/UPLC spectrophotometry procedure described in the OECD TG 492B. That allows reliable quantification of FD even when colored test items are not compatible with the standard absorbance measurement due to strong interference at a wavelength of 570±30 nm. This enables a meaningful and compliant evaluation for strongly colored test items.
